A simple method for the enrichment of phosphopeptides in urine sample is presented. Phosphopeptides were isolated with a C18 solid-phase and then concentrated in reversed-phase (RP) chromatography mode with a polymeric strong cation exchange (SPE) sorbent, enabling the subsequent protein digestion for subsequent identification by tandem mass spectrometry (MS/MS). The SPE enrichment combined with RP-LC-MS/MS improved phosphopeptide identification from urine sample in comparison with the direct analysis of the urine sample without enrichment. The method provided low sample consumption, high selectivity, and acceptable cost. Two commercial control human urine samples were analyzed with the proposed protocol and the results were compared with those from the previous study. The amount of phosphopeptides identified by the traditional method was dramatically lower than that identified with the affinity-based method. The number of identified phosphopeptides from the affinity-based method was about 10 times that from the traditional method.
